One of the complications that occur in DM sufferers is the occurrence of chronic complications. Hydrogel as a wound dressing has excellent biocompatibility. Human amnion membrane contains ECM with components such as growth factors, collagen, and proteoglycans as wound healing and tissue regeneration. To analyze the effect of hAM hydrogel treatment on the wound closure process in mice macroscopically and the area of collagen microscopically on days 3, 7, 14 and 21. The design of this research is experimental. Four treatment groups were used, namely DM NT, Sehat NT, DM carbopol and DM hAM. Wound measurements were carried out on days 3, 7, 14, 21 (macroscopic), tissue processing, Masson's trichrome staining, collagen area measurements (microscopic), and twoway anova test analysis was carried out using Graphpad Prism 10.2.3. The DM hAMD group experienced rapid wound closure and had the highest collagen area compared to the DM NT, Sehat NT, DM carbopol groups with a significant value of <0.05. Our findings suggest a promising potential of human amnion membrane hydrogel in influencing the healing process of diabetes mellitus wounds in vivo, as measured by the collagen area using ImageJ software.
